雖然有時把豚鼠歸類于小型嚙齒動物,但是,豚鼠的血脂代謝與大鼠、小鼠等差別較大,非常接近人類,用降血脂藥物干預時,豚鼠的血脂也表現為類似人類的反應。因此,豚鼠被常用于動脈粥樣硬化研究或者造模。

一、豚鼠動脈粥樣硬化模型造模的特點


豚鼠用于動脈粥樣硬化研究的優點:

(1)豚鼠對模型飼料中膽固醇敏感,能夠發生動脈粥樣硬化。

(2)豚鼠的血脂對模型飼料中脂肪類型或不同脂肪酸比例比較敏感,高脂高膽固醇模型飼料容易誘發動脈粥樣硬化。

(3)豚鼠用高膽固醇模型飼料或者高脂高膽固醇模型飼料喂養幾個月后能夠到達動脈粥樣硬化的斑塊階段(泡沫細胞,脂質沉積,水腫,等)。

(4)豚鼠與人類相似,需要依靠維生素C補充,有利于研究維生素C與動脈粥樣硬化發生和發展的關系。

(5)豚鼠動脈粥樣硬化模型適合降血脂和抗動脈粥樣硬化藥物活性的篩選和作用機制的研究。

豚鼠用于動脈粥樣硬化研究的缺點:

動脈粥樣硬化難以達到高級階段。

二、豚鼠動脈粥樣硬化造模的難點和動物要求


豚鼠對飼料中多種成分敏感甚至很敏感,無論是對照飼料還是模型飼料的配制,都是對飼料配比的考驗。

豚鼠對飼料中成分的敏感性到達什么程度呢?飼料中蛋白質、碳水化合物、脂肪三者的含量,脂肪類型和不同脂肪酸的比例,維生素C的含量,等等,都決定著豚鼠血漿脂蛋白、甘油三酯和膽固醇濃度,因此:

(1)豚鼠的對照飼料必須科學配制。

(2)豚鼠動脈粥樣硬化模型飼料必須嚴格控制和科學配比各項成分。

除了對照飼料和模型飼料,豚鼠本身的性別和年齡也決定動脈粥樣硬化的發生和發展。一般來說,應當優先使用雄性、成年或接近成年的豚鼠。

三、豚鼠動脈粥樣硬化模型飼料


豚鼠動脈粥樣硬化模型飼料包括:高膽固醇模型飼料,高脂高膽固醇模型飼料。

1.豚鼠動脈粥樣硬化高膽固醇模型飼料

豚鼠高膽固醇模型飼料中膽固醇含量必須達到一定高度,但是,過高可以導致某些器官(例如,腎臟)損傷。

用于豚鼠動脈粥樣硬化模型的高膽固醇模型,包括兩類:日糧型和純化型。

日糧型的價格較低,但純化型更適合豚鼠動脈粥樣硬化模型。下表列出的是南通特洛菲飼料科技有限公司提供的豚鼠動脈粥樣硬化高膽固醇模型飼料
